Sirtuins (PfSIR2A and PfSIR2B) are implicated to play pivotal roles in the silencing of sub-telomeric genes maintenance telomere length P. falciparum 3D7 strain. Here, we identify key factors that regulate cellular abundance activity these two histone deacetylases. Our results demonstrate PfSIR2A PfSIR2B transcriptionally downregulated at mid-ring stage response febrile temperature. We found molecular chaperone PfHsp90 acts as a repressor & B transcription. By virtue its presence promoter proximal regions helps recruiting H3K9me3, conferring heterochromatic state, thereby leading downregulation Such transcriptional can be reversed by addition 17-(allylamino)-17-demethoxygeldanamycin or Radicicol, potent inhibitors PfHsp90. The reduced occupancy PfSir2 var promoters leads de-repression genes. Thus, here uncover how exposure temperature, hallmark malaria, enables parasites manipulate expression prominent epigenetic modifiers PfSir2A PfSir2B.
